چکیده

Modern automated microscopes collect digital images at an astonishing pace. Automated image analysis can measure biological phenotypes quantitatively and reliably, and has therefore become a powerful tool for probing a wide variety of biological questions using microscopy. In this tutorial, we acquaint biologists with this important computational field and introduce some basic principles of image analysis, using typical strategies for twodimensional images of cultured cells in high-throughput screens as the primary example.

Stochastic optical reconstruction microscopy–based relative localization analysis (STORM-RLA) for quantitative nanoscale assessment of spatial protein organization

The spatial association between proteins is crucial to understanding how they function in biological systems. Colocalization analysis of fluorescence microscopy images is widely used to assess this. However, colocalization analysis performed on two-dimensional images with diffraction-limited resolution merely indicates that the proteins are within 200-300 nm of each other in the xy-plane and wi...

Extended Field Laser Confocal Microscopy (EFLCM): Combining automated Gigapixel image capture with in silico virtual microscopy

BACKGROUND Confocal laser scanning microscopy has revolutionized cell biology. However, the technique has major limitations in speed and sensitivity due to the fact that a single laser beam scans the sample, allowing only a few microseconds signal collection for each pixel. This limitation has been overcome by the introduction of parallel beam illumination techniques in combination with cold CC...

In vitro Delivery of HIV-1 Nef Antigen by Histidine-rich nona-arginine and Latarcin 1 peptide

Introduction: The Nef accessory protein is an attractive antigenic candidate in the development of HIV-1 DNA- or protein-based vaccines. The most crucial disadvantage of DNA and protein-based vaccines is their low immunogenicity, which can be improved by cell-penetrating peptides (CPPs) as effective carrier molecules. Methods: In this study, the HIV-1 Nef protein was generated in the Escherichi...

A Novel Spot-Enhancement Anisotropic Diffusion Method for the Improvement of Segmentation in Two-dimensional Gel Electrophoresis Images, Based on the Watershed Transform Algorithm

Introduction Two-dimensional gel electrophoresis (2DGE) is a powerful technique in proteomics for protein separation. In this technique, spot segmentation is an essential stage, which can be challenging due to problems such as overlapping spots, streaks, artifacts and noise. Watershed transform is one of the common methods for image segmentation. Nevertheless, in 2DGE image segmentation, the no...

Comparison of Two Quantitative Susceptibility Mapping Measurement Methods Used For Anatomical Localization of the Iron-Incorporated Deep Brain Nuclei

Introduction Quantitative susceptibility mapping (QSM) is a new contrast mechanism in magnetic resonance imaging (MRI). The images produced by the QSM enable researchers and clinicians to easily localize specific structures of the brain, such as deep brain nuclei. These nuclei are targets in many clinical applications and therefore their easy localization is a must. In this study, we aimed to i...
